**Broccoli sprout extract** is an orally administered botanical dietary-supplement preparation investigated as a \"green chemoprevention\" strategy for tobacco-related head and neck squamous cell carcinoma. The relevant clinical program led by Julie E. Bauman evaluated broccoli seed and sprout extract preparations, including the commercial product Avmacol, in healthy volunteers and in patients previously treated with tobacco-related head and neck squamous cell carcinoma. These preparations provide glucoraphanin and, in some formulations, active myrosinase; myrosinase converts glucoraphanin to **sulforaphane**, the principal bioactive isothiocyanate. Sulforaphane functionally activates the KEAP1-NRF2 cytoprotective-response pathway, increasing expression of phase II detoxification and antioxidant-response genes such as NQO1, glutathione S-transferases, and aldo-keto reductases. The HNSCC study was a completed phase 0 pharmacodynamic and bioavailability study, not a demonstrated recurrence-prevention efficacy trial.
