Overview

This is a combination of three monoclonal antibody drugs—brodalumab, ixekizumab, and secukinumab—all of which are interleukin-17 (IL-17) pathway inhibitors. Each drug targets the IL-17 axis but with slightly different mechanisms: - Brodalumab is a human monoclonal antibody that binds to the IL-17 receptor A (IL-17RA), blocking the activity of multiple IL-17 cytokines including IL-17A, IL-17F, and others. - Ixekizumab and secukinumab are both humanized monoclonal antibodies that specifically bind to and neutralize interleukin 17A (IL‑17A), preventing it from binding to its receptor. These agents are primarily used for moderate-to-severe plaque psoriasis and psoriatic arthritis. They have demonstrated high efficacy in reducing skin lesions and joint symptoms in clinical trials. The combination of these three drugs as a single therapy is not standard practice or approved; rather, each is typically used as monotherapy or sometimes in combination with conventional disease-modifying antirheumatic drugs (cDMARDs). There is limited evidence on combining multiple biologic agents targeting the same pathway due to potential safety concerns[2][1][5].
