Overview

BrP-01096 is a small molecule dual orexin receptor antagonist (DORA) being developed by Brisun Pharmaceuticals (Nantong) Co., Ltd. for the treatment of insomnia. It functions by blocking the binding of wake-promoting neuropeptides, orexin A and orexin B, to their receptors, Hypocretin receptor 1 (OX1R) and Hypocretin receptor 2 (OX2R). By inhibiting these receptors, BrP-01096 suppresses the drive to stay awake, thereby facilitating sleep onset and maintenance. Unlike traditional sedative-hypnotics that act on the GABA system, DORAs target the wakefulness-regulating system more specifically, which may offer a more favorable safety profile with reduced risks of next-day residual effects, cognitive impairment, or physical dependence. BrP-01096 is currently in Phase 2 clinical development for chronic insomnia.
