Overview

Bryolat is a nanoparticle-based therapy developed by Aphios International for the treatment of HIV-1 latency. It is designed to function as a latency-reversing agent (LRA) as part of a 'shock and kill' strategy aimed at achieving a functional cure for HIV. The drug utilizes bryostatin-1 (or a bryostatin analog), encapsulated in polymer nanospheres using Aphios's proprietary PNC (Polymer Nanospheres/Nanocapsules) technology. The mechanism of action involves the activation of Protein Kinase C (PKC), which induces the expression of latent HIV-1 proviruses in resting CD4+ T cells. This activation makes the infected cells visible to the immune system and susceptible to clearance by antiretroviral therapy or immune-mediated mechanisms. The nanoparticle formulation is intended to enhance the therapeutic index of the bryostatin compound by improving its stability, solubility, and targeted delivery while potentially reducing systemic toxicity.
