Overview

Bryophyllum pinnata fraction F4 is a research-stage botanical extract derived from the leaves of *Bryophyllum pinnata* (Lam.) Oken. It is obtained through chromatographic separation using a petroleum ether and ethyl acetate gradient (50:50 ratio). Phytochemical analysis indicates that this fraction is rich in steroidal glycosides, alkaloids, and steroids, with activity profiles resembling Bryophyllin A. In preclinical studies, fraction F4 has demonstrated significant growth inhibitory effects on human cervical cancer (HeLa) cells, with an IC50 of 91 μg/ml. Its mechanism of action involves the induction of apoptosis via the mitochondrial pathway, characterized by the upregulation of the pro-apoptotic protein Bax, downregulation of the anti-apoptotic protein Bcl-2, and subsequent activation of caspase-3 and cleavage of PARP-1. Furthermore, it exhibits anti-Human Papillomavirus (HPV) activity by suppressing the expression of oncogenic proteins c-Fos and c-Jun and inhibiting AP-1-specific DNA binding activity.
