bryostatin-1 + paclitaxel

A combination therapy consisting of bryostatin-1, a macrocyclic lactone that modulates protein kinase C (PKC), and paclitaxel, a microtubule-stabilizing chemotherapeutic agent. This combination was developed based on preclinical evidence suggesting synergistic antitumor effects. The sequence of administration appears critical, with paclitaxel followed by bryostatin-1 showing better efficacy than the reverse order. The combination has been studied in various cancer types including non-small cell lung cancer, pancreatic cancer, prostate cancer, and esophageal/stomach cancer, though clinical trials have shown limited efficacy and notable toxicity, particularly myalgia.
