Overview

BRZ-101 is an investigational inhaled formulation of bumetanide, a potent loop diuretic, being developed by Breeze Therapeutics for the prevention of bronchopulmonary dysplasia (BPD) in extremely low birth weight neonates. By delivering the drug directly to the lungs via nebulization, BRZ-101 aims to reduce pulmonary edema and improve lung compliance while avoiding the systemic electrolyte imbalances and ototoxicity often associated with systemic diuretic use. The mechanism of action centers on the inhibition of the Na-K-2Cl cotransporter (NKCC1), which is highly expressed in the neonatal lung and plays a critical role in fluid transport across the airway epithelium. By inhibiting NKCC1, BRZ-101 prevents the excessive accumulation of fluid in the alveolar spaces, thereby addressing one of the primary pathophysiological drivers of BPD in preterm infants.
