BSA01 is a fully human bispecific antibody-drug conjugate (ADC) targeting the epidermal growth factor receptor (EGFR) and the C-terminal subunit of mucin 1 (MUC1-C). Developed by Biocytogen using the RenLite humanized common light chain mouse platform, BSA01 is designed to treat solid tumors that highly co-express these two antigens, such as non-small cell lung cancer (NSCLC), esophageal squamous cell carcinoma, and triple-negative breast cancer. The ADC consists of a bispecific antibody conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE). BSA01 is engineered to target the membrane-bound MUC1-C cleavage product, providing better access to the tumor antigen in situ. Additionally, the EGFR-binding arm is optimized for affinity and internalization to enhance tumor selectivity and minimize potential skin toxicity associated with basal EGFR expression in healthy tissues.
