BsADC is a novel bispecific antibody-drug conjugate (ADC) designed to target the immune checkpoints Programmed death-ligand 1 (PD-L1) and B7-H3. This therapeutic agent combines the precise targeting capabilities of a bispecific antibody with the potent cytotoxic action of the small-molecule drug monomethyl auristatin E (MMAE). The drug aims to enhance antitumor efficacy and promote immune-mediated antitumor responses by selectively delivering its cytotoxic payload to cancer cells expressing these targets, thereby minimizing systemic toxicity and potentially overcoming limitations such as poor internalization and drug resistance associated with traditional ADCs. Studies have demonstrated its superior in vitro and in vivo tumor cytotoxicity compared to single-target ADCs or bispecific antibodies alone, leading to significant activation of tumor-specific immunity and induction of immunogenic cell death.
