Drug intelligence / Profile preview
BSBI-67003 is an orally available small molecule drug candidate under preclinical development for the treatment of yellow fever. Developed by the Baruch S. Blumberg Institute, the compound directly targets the yellow fever virus (YFV) nonstructural protein 4B (NS4B). By binding to NS4B, BSBI-67003 disrupts the structural integrity of endoplasmic reticulum-derived viral replication organelles (ROs) in infected cells, causing viral replication intermediates to leak into the cytoplasm and rapidly inhibiting nascent YFV RNA synthesis. Preclinical studies in hamster models have demonstrated potent, rapid-acting, and potentially Cmax-driven in vivo efficacy, providing significant protection against lethal infection even when administered at the peak of viremia.
