Overview

bscEphA2xCD3 is a bispecific T-cell engager (BiTE) antibody construct designed to target the Ephrin type-A receptor 2 (EphA2) and the CD3 epsilon subunit of the T-cell receptor complex. Developed using the BiTE technology platform, this molecule consists of two single-chain variable fragments (scFvs) linked in tandem. One scFv specifically binds to EphA2, a receptor tyrosine kinase that is highly overexpressed in a variety of solid tumors—including non-small cell lung cancer, breast cancer, and prostate cancer—while being minimally expressed in normal adult tissues. The second scFv binds to CD3 on T cells. By cross-linking tumor cells with cytotoxic T cells, bscEphA2xCD3 induces the formation of a cytolytic synapse, leading to the release of perforin and granzymes, which results in the MHC-independent apoptosis of the EphA2-expressing cancer cells. This mechanism allows for potent and selective tumor cell lysis regardless of the T-cell receptor specificity of the recruited T cells.
