BSI-302 is a first-in-class bispecific antibody-drug conjugate (BsADC) that simultaneously targets Trophoblast cell-surface antigen 2 (TROP2) and Epidermal Growth Factor Receptor (EGFR). Developed by Biosion using its proprietary SynTracer™ high-throughput endocytosis platform, the molecule is engineered to overcome resistance mechanisms associated with monotherapy ADCs by dual-targeting two highly expressed antigens in epithelial-derived solid tumors. By binding to both TROP2 and EGFR, BSI-302 is designed to enhance tumor selectivity and internalization, delivering a potent topoisomerase I inhibitor payload directly into malignant cells. This dual-targeting approach aims to improve therapeutic efficacy in cancers that may have heterogeneous antigen expression or have developed resistance to single-target therapies. BSI-302 is currently being investigated in Phase 1 clinical trials for the treatment of advanced solid tumors, including non-small cell lung cancer (NSCLC) and colorectal cancer.
