**BsNb PX-4** is a preclinical dual-targeting nanobody developed by researchers at Jecho Laboratories and Shanghai Jiao Tong University for pancreatic ductal adenocarcinoma. It is a single recombinant bispecific molecular entity comprising anti-PD-L1 and anti-CXCR4 nanobody domains linked by a flexible (G4S)3 peptide linker. The molecule blocks PD-L1-mediated immune-checkpoint signaling and inhibits CXCL12-mediated CXCR4 signaling, with reported effects including enhanced antitumor T-cell activity, reduced pancreatic stellate-cell activation, reduced cancer-associated fibroblast activity, delayed epithelial-mesenchymal transition, and improved T-cell infiltration into pancreatic tumors. In preclinical pancreatic cancer models, it showed antitumor activity in vitro and in xenografts.
