CM-14 (bstrazumab) is a preclinical-stage antibody-drug conjugate (ADC) developed by Curemeta for the treatment of solid tumors, including metastatic colon, gastric, and pancreatic cancers. It consists of a humanized IgG1 monoclonal antibody targeting the oncofetal antigen TRA-1-60 (an epitope on podocalyxin-like protein 1), conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a cathepsin-cleavable valine-citrulline dipeptide linker. By targeting TRA-1-60, which is expressed on pluripotent stem cells and various cancer cells but absent in most normal adult tissues, CM-14 aims to selectively deliver cytotoxic payloads to tumor cells. It is the humanized successor to the chimeric antibody CM-09.
