BTH1704 is a humanized IgG1 monoclonal antibody designed for intravenous administration. It targets aberrantly glycosylated Mucin 1 (MUC1), a protein overexpressed and abnormally glycosylated in various cancers. The drug exerts its antitumor effects primarily through antibody-dependent cell cytotoxicity and immunostimulation by binding to MUC1 on the surface of cancer cells, leading to opsonization and immune-mediated destruction of tumor cells. Originally based on the murine monoclonal antibody HMFG1, BTH1704 was developed as an antineoplastic agent for solid tumors such as breast and pancreatic cancer. Its development has been discontinued for breast cancer after clinical trials failed to progress beyond early phases; phase 1 trials in pancreatic cancer were also terminated[1][2][7].
