Aethon Therapeutics is developing a preclinical bispecific T cell engager (BiTE) using its proprietary HapImmune™ platform to target BTK-driven cancers, primarily hematologic malignancies. The therapy targets synthetic neoantigens (haptens) formed when covalent BTK inhibitors bind to their target protein within cancer cells. These drug-peptide conjugates are subsequently processed and presented on the cell surface by Major Histocompatibility Complex (MHC) molecules. The bispecific antibody is designed to recognize these specific peptide-MHC complexes and simultaneously engage CD3 on T cells, thereby recruiting the immune system to eliminate tumor cells, including those that have developed resistance to standard BTK inhibitor therapy. The program is backed by Apple Tree Partners and utilizes technology licensed from NYU Langone Health.
