Drug intelligence / Profile preview
BTK SH2 domain inhibitor is a preclinical-stage small molecule program being developed by Recludix Pharma for the treatment of autoimmune and inflammatory diseases, including multiple sclerosis (MS) and chronic spontaneous urticaria (CSU). Unlike traditional Bruton's tyrosine kinase (BTK) inhibitors that target the kinase domain, this first-in-class therapeutic approach targets the SH2 domain of BTK. By inhibiting the SH2 domain, the drug disrupts the formation of the pro-inflammatory signaling complex and blocks the scaffolding function of BTK. This mechanism offers exquisite selectivity over other kinases (such as TEC) and the broader SH2ome, potentially avoiding off-target toxicities like bleeding and liver signals associated with traditional BTK tyrosine kinase inhibitors (TKIs).
