BTM-3566 is a novel, orally available small molecule based on a pyrazolothiazol-backbone, developed by Bantam Pharmaceutical for the treatment of various cancers, with an initial clinical focus on mature B-cell lymphomas such as diffuse large B-cell lymphoma (DLBCL), mantle cell lymphoma (MCL), and follicular lymphoma (FL)[1][2][3][4]. The drug demonstrates potent anti-cancer activity in preclinical models, including those resistant to standard therapies like CAR-T cell therapy[1]. Mechanistically, BTM-3566 disrupts mitochondrial function in tumor cells and activates the ATF4-integrated stress response (ISR) pathway via the mitochondrial protease OMA1. This leads to phosphorylation of eukaryotic translation initiation factor 2α (eIF2α) through activation of HRI kinase, resulting in apoptosis and tumor regression[2][5]. The compound is currently being evaluated in Phase 1 clinical trials for relapsed/refractory mature B-cell lymphomas[1][3].
