Drug intelligence / Profile preview
BTPA-Ma is a mitochondria-targeted, triphenylamine-bridged bis-boronate small molecule designed for the dual-mode treatment of bladder cancer [P0657]. Developed by Shandong University and Qilu Hospital of Shandong University, BTPA-Ma exhibits context-dependent, bidirectional regulation of reactive oxygen species (ROS) [P0657]. Under white light irradiation, BTPA-Ma acts as a photosensitizer, triggering a massive ROS storm that causes mitochondrial rupture, endoplasmic reticulum stress via the PERK-EIF2α pathway, and immunogenic cell death (ICD) [P0657]. In the absence of light (dark conditions), BTPA-Ma functions as a ROS scavenger, gradually consuming intrinsic ROS in tumor cells, which upregulates PTEN and inhibits the PI3K-AKT proliferative pathway [P0657]. This dual functionality allows BTPA-Ma to suppress tumor growth and remodel the immunosuppressive tumor microenvironment [P0657].
