BTRP011L is a potent and selective molecular glue degrader targeting the G1 to S phase transition 1 (GSPT1) protein. Developed by BioTheryX, the compound is designed to induce the degradation of GSPT1, a translation termination factor that is often overexpressed in various cancers and is essential for the survival of certain tumor cells. By leveraging the ubiquitin-proteasome system, BTRP011L promotes the recruitment of GSPT1 to the CRL4-CRBN E3 ubiquitin ligase complex, leading to its ubiquitination and subsequent proteasomal degradation. This mechanism results in the inhibition of protein synthesis and induction of apoptosis in cancer cells. BTRP011L is primarily being investigated for the treatment of hematological malignancies, including acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS).
