Overview

BTSC mRNA-loaded DCs is an autologous, personalized dendritic cell vaccine designed for the treatment of recurrent glioblastoma multiforme (GBM). Developed by John Sampson and colleagues at Duke University, the therapy involves harvesting a patient's own dendritic cells and loading them with messenger RNA (mRNA) derived from brain tumor stem cells (BTSCs), specifically those expressing the CD133 marker. These stem cells are believed to be responsible for tumor recurrence and resistance to standard therapies. By presenting BTSC-specific antigens, the vaccine aims to prime the patient's immune system to recognize and eliminate the tumor-propagating cell population. In cases where specific BTSC RNA cannot be isolated in sufficient quantities, total tumor RNA (ttRNA) is utilized as an alternative antigen source.
