Overview

Buccolam is a buccal formulation of midazolam, a benzodiazepine small molecule developed by Takeda for use in Japan. It is specifically formulated as an oromucosal solution for the treatment of status epilepticus and prolonged acute convulsive seizures. Midazolam functions as a positive allosteric modulator of the GABA-A receptor, which enhances the inhibitory effects of GABA in the brain, thereby reducing neuronal excitability and terminating seizure activity. Takeda has conducted special drug use surveillance in Japan to evaluate its safety and efficacy in patients with non-convulsive status epilepticus (NCSE) in community settings, following its approval in Japan in September 2020.
