Bud-ADC is an experimental antibody-drug conjugate (ADC) consisting of a cross-species anti-CD74 monoclonal antibody conjugated to the potent glucocorticoid budesonide via a cleavable linker. It is designed to selectively deliver budesonide to CD74-expressing immune cells, such as B cells and dendritic cells, which are central to the pathogenesis of systemic lupus erythematosus (SLE). By concentrating the glucocorticoid within target immune cells, Bud-ADC aims to achieve robust immunosuppression while mitigating the systemic toxicities associated with conventional glucocorticoid therapy. Preclinical studies in SLE mouse models have demonstrated that Bud-ADC effectively reduces autoantibody levels, splenomegaly, and renal damage, showing superior efficacy compared to systemic budesonide administration.
