Budoprutug is an Fc-engineered, anti-CD19 targeted monoclonal antibody developed for the treatment of immune-mediated diseases. It works by targeting the CD19 antigen on B cells, leading to antibody-dependent cell cytotoxicity and depletion of both peripheral and tissue-resident B cells. This mechanism aims to reduce pathological B-cell activity in diseases such as membranous glomerulonephritis, systemic lupus erythematosus (SLE), and idiopathic thrombocytopenic purpura. Budoprutug has demonstrated dose-dependent B-cell depletion in preclinical models and clinical trials, with promising Phase 1b results showing remission in patients with primary membranous nephropathy (pMN). The drug is being developed by Climb Bio, which acquired it from Tenet Medicines. Budoprutug is also being formulated for subcutaneous administration to allow low volume injections while maintaining efficacy.
