Buflomedil

Buflomedil is a vasoactive small molecule drug primarily used to treat symptoms of peripheral arterial occlusive disease (PAOD), such as intermittent claudication. It acts by increasing blood flow to the brain and other parts of the body through vasodilation. The mechanism of action involves inhibition of alpha-adrenergic receptors (both α1 and α2), inhibition of phosphodiesterase enzymes, weak calcium antagonistic effects, improved erythrocyte deformability, inhibition of platelet aggregation, and oxygen-sparing activity. Buflomedil was developed in France (notably by Laboratoire Lafon) and has been marketed under various trade names including Loftyl and Fonzylane. It was widely used in Europe from the 1970s but has since had its marketing authorizations suspended throughout the EU due to concerns over severe neurological and cardiac toxicity[1][4][5][6][8].
