Overview

Buformin is an oral antidiabetic drug of the biguanide class, chemically related to metformin and phenformin. It acts as an antihyperglycemic agent by delaying glucose absorption from the gastrointestinal tract, increasing insulin sensitivity and glucose uptake into cells, and inhibiting hepatic gluconeogenesis. Buformin does not stimulate insulin release and does not cause hypoglycemia in non-diabetics. Its mechanism involves activation of AMP-activated protein kinase (AMPK), which regulates cellular energy homeostasis, and may also involve inhibition of TNF-alpha signaling. Buformin was introduced in 1958 for type 2 diabetes mellitus treatment but has been withdrawn from most markets due to a high risk of lactic acidosis[1][3][4][7].
