Drug intelligence / Profile preview
Bumetanide nasal spray (RSQ-777) is a novel intranasal formulation of the loop diuretic bumetanide, developed as an alternative to oral and intravenous forms for the treatment of edema associated with congestive heart failure, hepatic disease, and renal disease. Bumetanide acts by inhibiting sodium and chloride reabsorption in the thick ascending limb of the loop of Henle in the kidney, promoting diuresis and natriuresis. The nasal spray formulation is designed for outpatient self-administration to address challenges such as diuretic resistance due to impaired gastrointestinal absorption in patients with fluid overload. Clinical trials have shown that RSQ-777 achieves similar urine output and blood concentrations compared to oral and IV formulations but with faster absorption (33% faster than oral), more consistent pharmacokinetics (lower variability), rapid onset of action for sodium excretion, and good tolerability. The product aims to reduce hospital admissions by enabling effective at-home management of fluid overload[1][2][3][4][5][6][7][8].
