Buntanetap (formerly known as Posiphen or ANVS-401) is an orally bioavailable small molecule being developed by Annovis Bio for the treatment of neurodegenerative diseases, including Alzheimer's and Parkinson's disease. It acts as a translational inhibitor that targets the mRNA of several neurotoxic proteins by binding to the iron-responsive element (IRE) in their 5' untranslated region (UTR). This mechanism specifically blocks the translation of amyloid precursor protein (APP), tau, alpha-synuclein, and TDP-43. By reducing the levels of these neurotoxic proteins, buntanetap aims to restore axonal transport, reduce neuroinflammation, and improve synaptic transmission. The drug is currently being evaluated in Phase 3 clinical trials for early Alzheimer's disease, focusing on both symptomatic improvement and disease-modifying potential.
