Overview

**Buparlisib + atezolizumab** is an investigational combination therapy composed of buparlisib, an oral pan-class I phosphoinositide 3-kinase (PI3K) inhibitor, and atezolizumab, a humanized monoclonal antibody that targets programmed death-ligand 1 (PD-L1). Buparlisib inhibits all isoforms of class I PI3K, resulting in cell cycle arrest and apoptosis in tumor cells. Atezolizumab blocks the interaction between PD-L1 and its receptors (PD-1 and B7.1), thereby enhancing antitumor T-cell activity. This combination is under clinical investigation for use in advanced/metastatic solid tumors, as part of a strategy to combine targeted therapy with immunotherapy[1].
