Overview
Buparlisib (BKM120) is an investigational, oral, pan-class I phosphatidylinositol 3-kinase (PI3K) inhibitor. This specific therapeutic combination, studied at the Roswell Park Cancer Institute, pairs buparlisib with the chemotherapy agents gemcitabine and cisplatin. Buparlisib targets the PI3K/AKT/mTOR signaling pathway, which is often dysregulated in cancer, while gemcitabine (a nucleoside analog) and cisplatin (a platinum-based agent) work to inhibit DNA synthesis and induce DNA damage. The combination was evaluated in a Phase I dose-escalation trial for advanced solid tumors, with a specific expansion cohort for non-small cell lung carcinoma (NSCLC).
