Buparlisib + paclitaxel is a combination therapy under investigation for the treatment of various cancers, most notably recurrent or metastatic head and neck squamous cell carcinoma (HNSCC) and HER2-positive breast cancer. Buparlisib is an orally bioavailable, highly selective pan-class I phosphatidylinositol 3-kinase (PI3K) inhibitor that targets all class I PI3K isoforms (p110α, β, γ, δ), thereby inhibiting the PI3K/AKT signaling pathway. This inhibition leads to reduced tumor cell proliferation, increased apoptosis, and decreased angiogenesis[6][8]. Paclitaxel is a well-established chemotherapeutic agent that stabilizes microtubules and inhibits cell division. The combination has shown synergistic antitumor activity in preclinical models and early-phase clinical trials[4]. Clinical studies have demonstrated improved outcomes compared to paclitaxel alone in patients with HNSCC who have progressed after prior therapies[1][2][5]. Buparlisib was originally developed by Novartis and later licensed to Adlai Nortye for further development[8][5].
