Drug intelligence / Profile preview
Buprenorphine (specifically the Sublocade formulation by Indivior) is an extended-release subcutaneous injection indicated for the treatment of moderate-to-severe opioid use disorder (OUD). It utilizes the ATRIGEL delivery system, which consists of a biodegradable poly (DL-lactide-co-glycolide) polymer dissolved in a biocompatible solvent that solidifies upon contact with body fluids to form a solid depot. This depot subsequently delivers buprenorphine over a one-month period. Mechanistically, buprenorphine acts as a partial agonist at the mu-opioid receptor and an antagonist at the kappa-opioid receptor. This pharmacological profile helps to reduce opioid cravings and withdrawal symptoms while exhibiting a ceiling effect on respiratory depression, providing a safer alternative to full opioid agonists. It is intended for use as part of a complete treatment program that includes counseling and psychosocial support.
