Bupropion + methylphenidate is a combination of two small molecule pharmaceuticals, each with distinct but complementary mechanisms of action. Bupropion is an atypical antidepressant that acts primarily as a norepinephrine–dopamine reuptake inhibitor (NDRI) and also antagonizes nicotinic acetylcholine receptors. It is commonly used for major depressive disorder, seasonal affective disorder, and smoking cessation[4][5]. Methylphenidate is a central nervous system stimulant that inhibits the reuptake of dopamine and norepinephrine by blocking their respective transporters, leading to increased concentrations in the synaptic cleft; it is primarily indicated for attention deficit hyperactivity disorder (ADHD) and narcolepsy[8]. The combination has been explored off-label for treatment-resistant depression or ADHD with comorbid mood disorders.
