Overview

Bupropion + methylphenidate is a combination of two small molecule pharmaceuticals, each with distinct but complementary mechanisms of action. Bupropion is an atypical antidepressant that acts primarily as a norepinephrine–dopamine reuptake inhibitor (NDRI) and also antagonizes nicotinic acetylcholine receptors. It is commonly used for major depressive disorder, seasonal affective disorder, and smoking cessation[4][5]. Methylphenidate is a central nervous system stimulant that inhibits the reuptake of dopamine and norepinephrine by blocking their respective transporters, leading to increased concentrations in the synaptic cleft; it is primarily indicated for attention deficit hyperactivity disorder (ADHD) and narcolepsy[8]. The combination has been explored off-label for treatment-resistant depression or ADHD with comorbid mood disorders. Both drugs increase dopaminergic and noradrenergic neurotransmission in the brain but through different transporter inhibition profiles. This combination may have additive effects on mood, motivation, attention, or energy levels but carries an increased risk of adverse events such as hypertension or seizures due to overlapping pharmacological actions[1][7].
