Overview
Burixafor is an orally bioavailable small molecule antagonist of the C-X-C chemokine receptor type 4 (CXCR4). By binding to CXCR4, it blocks the interaction between this receptor and its ligand stromal derived factor 1 (SDF-1 or CXCL12), thereby inhibiting receptor activation. This mechanism leads to rapid mobilization of hematopoietic stem and progenitor cells from the bone marrow into peripheral blood. Burixafor is being developed primarily as a first-in-class agent for stem cell mobilization in patients undergoing hematopoietic stem cell transplantation, particularly those with hematological malignancies such as multiple myeloma, Hodgkin's disease, and non-Hodgkin's lymphoma. The drug was originally developed by TaiGen Biotechnology and is also under development by GPCR Therapeutics[1][3][5][6][7].
