Overview

Buserelin is a synthetic nonapeptide analogue of gonadotropin-releasing hormone (GnRH, also known as luteinizing hormone-releasing hormone, LHRH). It acts as a superagonist at the GnRH receptor, initially stimulating the release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), which leads to increased production of sex hormones. However, with chronic administration, it causes desensitization and downregulation of pituitary GnRH receptors, resulting in suppression of LH and FSH secretion. This leads to decreased production of testosterone in men and estrogen/progesterone in women. Buserelin is primarily used for the treatment of prostate cancer and endometriosis; it is also indicated for premenopausal breast cancer, uterine fibroids, central precocious puberty, assisted reproduction protocols for female infertility, and sometimes as part of transgender hormone therapy. It is administered via subcutaneous injection or nasal spray[1][3][4][5].
