Overview

Buspirone + pindolol is a combination of two small molecule drugs, buspirone and pindolol, investigated primarily for their effects on serotonergic neurotransmission and as an augmentation strategy in the treatment of major depressive disorder. Buspirone is a serotonin 5-HT1A receptor partial agonist with anxiolytic and antidepressant properties, while pindolol acts as a nonselective beta blocker with intrinsic sympathomimetic activity and also exhibits antagonistic activity at postsynaptic 5-HT1A receptors. The rationale for combining these agents is to preferentially activate postsynaptic 5-HT1A receptors (via buspirone) while blocking presynaptic autoreceptors (via pindolol), potentially accelerating or enhancing antidepressant response. Clinical studies have shown that this combination can induce rapid and robust reductions in depressive symptoms, likely through modulation of serotonergic signaling[2][3][5]. EEG studies suggest both drugs individually—and in combination—produce similar shifts in EEG frequency spectrum mediated by somatodendritic 5-HT1A receptors[1][6].
