The regimen consists of the alkylating agent busulfan and the purine analog fludarabine, combined with low-dose total body irradiation (TBI). This combination serves to suppress the recipient's immune system and deplete bone marrow niches, facilitating the successful engraftment of infused donor-derived hematopoietic stem cells while minimizing the high toxicity typically associated with standard myeloablative conditioning.
