Overview

Butamben is a local anesthetic of the ester type used primarily for topical anesthesia of mucous membranes (excluding the eyes). It is chemically known as n-butyl-p-aminobenzoate and acts by inhibiting voltage-gated calcium channels (including T-type and N-type) and sodium channels in dorsal root ganglion neurons. This results in reduced electrical excitability and selective inhibition of pain signal transmission through C fibers without significant loss of motor function. Due to its low water solubility, it is suitable only for topical use. It is commonly found as a component in multi-agent topical anesthetics such as Cetacaine. The FDA has removed parenteral forms from the market due to poor solubility[1][2][4][5][6].
