Overview

Butriptyline is a tricyclic antidepressant (TCA) that has been used in Europe since 1974 for the treatment of depression. It is structurally related to amitriptyline, differing by an isobutyl side chain. The drug acts primarily by inhibiting the reuptake of serotonin and norepinephrine in the brain, thereby increasing their synaptic availability and improving mood. Butriptyline also exhibits potent antihistamine and anticholinergic effects, leading to sedation and typical TCA side effects such as dry mouth and constipation. Unlike some other TCAs, it has relatively weak alpha-1 adrenergic blocking activity and little effect on norepinephrine reuptake[1][2][3][6]. Its primary indication was unipolar depression.
