Butyltriphenylphosphonium bromide (TP187) is a mitochondriotropic triphenylphosphonium (TPP) salt with broad-spectrum antineoplastic and anti-angiogenic activity. Discovered through a high-throughput screen of drug-like small molecules, TP187 selectively accumulates in the mitochondria of cancer cells driven by the mitochondrial membrane potential. Upon uptake, it disrupts mitochondrial function, leading to decreased oxygen consumption, increased mitochondrial superoxide production, and cell cycle arrest independent of p53 status. In preclinical models, TP187 has demonstrated significant inhibition of tumor growth and angiogenesis, particularly in breast and pancreatic cancer models, without causing systemic toxicity.
