Overview

**BW-20507** is a double-stranded, N-acetylgalactosamine (GalNAc)-conjugated small interfering RNA (siRNA) specifically designed to target the S region of hepatitis B virus (HBV) messenger RNA in hepatocytes using hepatocyte-specific delivery via asialoglycoprotein receptor-mediated uptake. Developed by Argo Biopharma, BW-20507 inhibits several HBV mRNA transcripts, broadly suppressing expression of viral antigens (except HBx) and resulting in a marked reduction of HBsAg and HBV DNA levels in both nucleos(t)ide (NUC)-experienced and NUC-naïve chronic hepatitis B (CHB) patients. Phase 1/2a studies show significant, dose-dependent reductions in HBsAg (up to -3.2 log₁₀ IU/mL for higher-dose cohorts), with some patients achieving HBsAg loss, as well as potent antiviral activity and a favorable safety profile. The drug has received Breakthrough Therapy Designation in China based on early clinical data and is being advanced into Phase 2b clinical development as monotherapy and in combination with pegylated interferon for CHB.
