BW284c51 is a potent and highly selective inhibitor of acetylcholinesterase (AChE). It is widely utilized as a research tool to differentiate the activity of AChE from butyrylcholinesterase (BChE), as it exhibits significantly higher affinity for the former. Recent academic research has investigated its potential role in modulating the endothelial marrow niche to accelerate hematopoietic recovery following hematopoietic cell transplantation (HCT). By inhibiting the clearance of acetylcholine, BW284c51 increases acetylcholine exposure, which has been shown in preclinical models to promote the proliferation of marrow endothelial cells and support the engraftment of hematopoietic stem cells (HSCs). This mechanism may be particularly relevant for recovering the HSC niche after endothelial injuries, such as transplant-associated thrombotic microangiopathy (TA-TMA).
