Overview

BWC0977 is a novel, broad-spectrum antibacterial agent developed to address multidrug-resistant (MDR) bacterial infections. It demonstrates significant efficacy against both Gram-positive and Gram-negative pathogens, including strains resistant to fluoroquinolones, carbapenems, and colistin[1][2][3][5]. Its mechanism of action involves inhibition of bacterial DNA replication by targeting both DNA gyrase and topoisomerase IV—enzymes essential for bacterial DNA processes[4][6][8]. Unlike quinolones that stabilize double-strand breaks, BWC0977 stabilizes single-stranded DNA breaks at the GyrA dimer interface and triggers an SOS response in bacteria[6]. The drug has completed Phase 1 clinical trials with demonstrated safety and dose-proportional pharmacokinetics in healthy volunteers[5][6], and is being advanced through further clinical development via a collaboration between Bugworks Research and the Global Antibiotic Research & Development Partnership (GARDP)[2][7]. An oral formulation is also under development to improve accessibility for patients[1][3].
