BWC4221 (also known as BWC3416) is a preclinical-stage small molecule inhibitor of methionine adenosyltransferase 2A (MAT2A) being developed by Bugworks Research. It is designed as a synthetic-lethal therapeutic strategy targeting methylthioadenosine phosphorylase (MTAP)-deleted cancers, which exhibit a dependency on MAT2A. MTAP deletion is a common genomic alteration in various malignancies, including pancreatic, lung, bladder, gastrointestinal cancers, glioblastoma, and mesothelioma. By inhibiting MAT2A, BWC4221 aims to selectively disrupt cancer cell survival while sparing normal tissues. The drug is currently in the lead generation phase of preclinical development.
