Overview

BX-912 is a small molecule inhibitor of 3-phosphoinositide-dependent protein kinase 1 (PDK1). Originally developed by Berlex Biosciences for oncology indications, it has more recently been identified as a potential therapeutic for pulmonary arterial hypertension (PAH). Research presented at ATS 2025 indicates that BX-912 can rescue impaired expression of the transcription factor SOX17, which is critical for pulmonary endothelial cell function. By restoring SOX17-regulated gene expression and pathways, BX-912 has demonstrated the ability to ameliorate pulmonary hypertension and reduce right ventricular systolic pressure in preclinical models. It is currently utilized primarily as a research tool compound to study PDK1 signaling and endothelial biology.
