Overview

BXL0097 is a synthetic "Gemini" analog of calcitriol (1,25-dihydroxyvitamin D3) that acts as an agonist of the Vitamin D Receptor (VDR). Gemini analogs are structurally unique in that they possess two side chains at the C21 position, allowing them to interact with the receptor in a way that induces a distinct conformational change and enhances biological activity compared to natural calcitriol. BXL0097 is specifically a 19-nor analog, meaning it lacks the exocyclic methylene group at the C19 position of the A-ring. In preclinical studies, BXL0097 has demonstrated anti-proliferative and tumor-reducing effects, particularly in breast cancer models, by activating VDR-mediated transcription, although it is reported to be less potent than its 19-methylene counterpart, BXL0124.
