BXQ-350 is a first-in-class biologic drug composed of a synthetic form of the human glycoprotein saposin C (SapC) and the phospholipid dioleoylphosphatidylserine sodium (DOPS), forming stable nanovesicles. It acts as a sphingolipid metabolism modulator and selectively targets tumor cell phosphatidylserine (PS) patches, which are characteristic of tumor cells and neovasculature. Upon binding to PS, BXQ-350 catalyzes acid sphingomyelinase activity, leading to increased ceramide production and subsequent induction of apoptosis in cancer cells. Preclinical studies have shown that BXQ-350 can cross the blood-brain barrier and induce cell death in various cancer types, including high-grade gliomas and pancreatic cancers. Clinically, it is being investigated for advanced solid tumors—including brain tumors—and chemotherapy-induced peripheral neuropathy (CIPN). The drug is under development by Bexion Pharmaceuticals[1][2][3][5][7][8].
