Drug intelligence / Profile preview
BYON14156 is a preclinical-stage, first-in-class antibody-drug conjugate (ADC) developed by Byondis. It is designed to target an undisclosed tumor antigen highly expressed across multiple solid tumor types. BYON14156 utilizes Byondis' proprietary antifolate payload platform, which features an optimized antifolate payload that potently inhibits dihydrofolate reductase (DHFR). The payload is conjugated via a glucuronide-based linker designed to improve the therapeutic index. BYON14156 is specifically engineered to overcome payload resistance, a major limitation of current topoisomerase-I and tubulin inhibitor-based ADCs, and has demonstrated robust in vitro and in vivo efficacy across various tumor models, including those resistant to standard ADC payloads. The drug is currently in IND-enabling studies, with clinical trial initiation anticipated in 2027.
