Overview

**BZD1901** is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting mesothelin (MSLN), engineered to secrete a PD-1 nanobody for autocrine blockade of the PD-1/PD-L1 axis, enhancing antitumor activity in immunosuppressive solid tumor microenvironments. Developed by **Shanghai Cell Therapy Group** using non-viral gene editing, it addresses CAR-T exhaustion and PD-L1-mediated resistance. It received **FDA Orphan Drug Designation** for mesothelioma and shows early clinical efficacy, including 1 CR and 6 PR in 11 malignant pleural mesothelioma (MPM) patients, with promising safety in phase I trials for MSLN-positive advanced solid tumors.[1][3][4][7]
